What Makes SS-31 Different to Make

SS-31 has the sequence D-Arg-Dmt-Lys-Phe-NH2, molecular formula C32H49N9O5, molecular weight 639.8 g/mol and CAS number 736992-21-5. That is four residues, ending in an amide rather than a free acid.

Two details set it apart. First, the arginine is the D form, the mirror image of the usual L amino acid. Second, Dmt stands for 2,6-dimethyl-tyrosine, a tyrosine carrying two extra methyl groups on its ring. Neither is one of the twenty common building blocks, so each has to be sourced as a specialised reagent. Therefore, a four-residue chain can still carry a raw-material cost that a longer, standard sequence does not.

Identity by Mass

Mass spectrometry compares the observed mass with the expected 639.8 g/mol for the free peptide. A missing residue, or a plain tyrosine in place of Dmt, shows up as a different mass, 28 units lighter in that second case. So identity testing, not purity alone, is what confirms you are paying for SS-31.

Why Salt Form Matters More on a Short Peptide

SS-31 carries three basic groups: the arginine side chain, the lysine side chain and the free N-terminus. Each can hold a counter-ion, so a vial of powder is never 100% peptide by weight.

The FDA label for the approved medicine shows the scale of this. It lists elamipretide as a hydrochloride salt, C32H49N9O5·3HCl, with a molecular weight of 749.2. In that form, about 85% of the weight is the peptide itself. Other salts weigh more: three acetate counter-ions would bring the peptide share to about 78%, and three trifluoroacetate counter-ions to about 65%.

What the Literature Studies

SS-31 belongs to a family of small aromatic-cationic peptides studied for how they reach mitochondria. A 2013 paper in the Journal of the American Society of Nephrology by Birk and colleagues reported that SS-31 binds cardiolipin, a phospholipid of the inner mitochondrial membrane, and studied it in vitro and in a rat model of kidney ischaemia.

The compound has also reached the clinic. On 19 September 2025, the US FDA granted accelerated approval to Forzinity (elamipretide), from Stealth BioTherapeutics, for Barth syndrome, a rare inherited mitochondrial disorder. Accelerated approval means continued approval depends on a confirmatory trial. That approval covers Forzinity as a finished medicine in the United States. Federal Decree-Law No. 38 of 2024 governs medical products and pharmaceutical establishments in the UAE, and it took effect on 2 January 2025. The Emirates Drug Establishment (EDE) is the federal body that acts on unapproved medicines. In July 2026, the EDE acted against 71 sources selling unapproved peptide products and referred 14 influencers to the National Media Office.

A “research use only” label describes presentation, not registration. A US approval for Forzinity does not change that for research material sold in the UAE.
